Missing Folders in Hard Drive ?

I have got an Intel iMac model with 2 GB RAM and 500 GB hard drive that is connected to my Airport Extreme. One of the folders located in the hard drive is acting abnormally. The folder is visible but when I am trying to see its contents, I am able to see the contents for a short duration and then they disappear. I did the following things to get the data recovered from the folder: * Used single user as well as verbose mode * Resetted the NVRAM * Used AHT from Tiger disc * Checked on the bootable devices In spite of doing all these things, I was not able to see the contents of the folder. Am I doing anything wrong with my procedure? Kindly help me out so that I am able to recover the contents from this inaccessible folder.
